在現今的食品當中,潛藏著許多對人體造成健康危害的毒素,這些毒素來源有:黴菌毒素、細菌類毒素、重金屬毒素、農藥和化學污染等等,對人體可是造成疾病的發生。其中黃麴毒素AFB1為黴菌毒素的一種,主要污染來源為豆類食品。在毒性上,相對於其它具有相似結構的黃麴毒素,被認為是毒性較強的黴菌毒素。從文獻上,也發現在真菌類當中,具有其降低AFB1毒性的解毒酵素,反應後發現在螢光下偵測其AFB1吸收值降低。因此在實驗研究中,也希望能從真菌類樟芝發酵中,找出具有可降低AFB1毒性的解毒酵素。
本研究以樟芝Antrodia camphorata BCRC 35396於氣舉式發酵槽培養下,找尋具有降低毒素毒性的解毒酵素,其反應上造成AFB1螢光降低,並將發酵液進行酵素分離純化。
解毒酵素的純化步驟是將發酵澄清液利用硫酸銨沉降、透析,再經過DEAE Sepharose陰離子交換管柱層析分離純化。最後由SDS-PAGE電泳分析,找出此解毒酵素分子量為54kDa。
在反應部份,則是將純化之酵素與AFB1反應,利用AFB1在螢光excitation 365nm偵測下,emission 420nm 會有吸收值,來作為判斷毒素反應變化情況。
