摘 要
桑黃為一種食藥用菇,早在數百年前,中國明代偉大的藥物家李時珍已將桑黃作為一味藥材收載於《本草綱目》中。在許多古書籍中也曾推崇其各種生理、免疫及治療功能,是中國人自古以來珍貴稀有的藥用真菌之一。一九六八年,日本國立癌症中心研究所及東京大學醫學部的研究團體,進行大量有關蕈類抗腫瘤性的實驗,其中包含大家從未曾聽過的『野生桑黃』。實驗結果發現,野生桑黃腫瘤抑制率高達96.7%而位居第一,因此桑黃可說是未來相當具有潛力開發成為抗癌藥物的藥用真菌之ㄧ。
本研究先利用搖瓶培養實驗,於既有的培養基中添加0、0.02、0.05、0.1、0.2、0.3%不同濃度之氯化鈉。實驗結果發現,添加氯化鈉會抑制菌體生長,隨著添加濃度愈高,菌體量愈少,菌體顆粒也愈小。然而氯化鈉的添加卻促使高分子多醣產生,其原因可能是添加的氯化鈉改變發酵液之滲透壓,促使菌體將胞內多醣釋放於發酵液中,也可能因為添加氯化鈉改變了菌體的代謝行為,促使多醣產生。在適當的添加濃度,隨著添加的氯化鈉濃度愈高,多醣產量愈多。當添加濃度高達0.3%,菌體完全不會生長。
在發酵槽實驗中,添加氯化鈉會促使多醣產生,其中以氯化鈉添加濃度0.1%,其多醣產量最高,為329mg/L。多醣為一次代謝產物,菌體量之多寡可能會間接影響其多醣產量,因此以血清瓶模擬發酵槽,做Fed-Batch實驗,於第一階段先讓菌體生長,在於適當時機添加氯化鈉,促使菌體生產多醣。初步結果顯示,Fed-Batch隨著氯化鈉濃度愈高,其單位菌體所產多醣量也愈高,幾乎呈線性正比關係,印證發酵槽Fed-Batch的可行性。
在改變供氧量對多醣產量影響方面,降低通氣量會使得碳源氧化速率變慢,因此影響菌體生長,當通氣量由0.5vvm降低為0.1vvm時,其菌體量也由11.0587g/L 降低到6.315g/L。雖然整體總多醣量並沒有因為通氣量降低而提升,但是單位菌體所產多醣量確實有提升效果,Yp/x由0.03(高通氣量0.5vvm)提升到0.048(低通氣量0.1vvm)。
